Four hurt in crash, attack at California Walmart

SAN JOSE, Calif. (AP) ? Four people were hurt Sunday when a driver crashed his car into a Wal-Mart and then assaulted customers inside, officials said.

The man hit two cars in the parking lot at about 11:15 a.m. with his red Oldsmobile Cutlass sedan then crashed through the storefront near the pharmacy and collided with a beer display before stopping, police and witnesses said.

The unidentified driver then got out of his car and used a blunt object to attack people, San Jose police Officer Albert Morales said. The driver was arrested when officers arrived.

Investigators have not determined how fast the driver ? described as a man in his 30s ? was going at the time of the crash but the car went about 20 feet into the Wal-Mart Supercenter that had about 70 people inside in San Jose, Morales said.

One person suffered what Morales described as serious injuries. He did not know the extent of the injuries to the three other people but said they were not life-threatening. The injured included a store employee.

Customer Sharon Kaye told the San Jose Mercury News the driver sideswiped her car as he made several runs around the parking lot before driving between poles at the entrance and crashing into the store.

"At first, I thought I may have done something to anger him while driving," she said. "But then I realized he was out to get into the store."

After the crash, the entrance to the Wal-Mart was roped off with yellow police tape, and workers put up large boards covering the automatic doors where the car had entered.

A Wal-Mart spokeswoman told the Mercury News that the store remained shut down for several hours, and an employee was among those hurt.

"We're obviously very concerned about the associate who was injured," spokeswoman Kory Lundberg said.

Shopper and witness Tianna Doan told the newspaper the employee was a cashier who was hit with the object and had a head injury.

Calls to the store by The Associated Press went unanswered Sunday.

Bill Allowing Gold and Silver as Money; Bureaucratic Nightmare ...

I have encouraging news in the state of Arizona where lawmakers back gold, silver as currency.

The measure is Arizona's latest jab at the federal government, which prohibits states from minting their own money. It also reflects a growing distrust of government-backed money.

The bill, which advanced in a 4-2 vote by a House committee Monday, states that gold and silver should be legal currency not subject to tax or regulation as property. The Republican-led Senate gave the bill its blessing in February in a 17-11 partisan vote.

The bill would let people use the precious metals as money as long as businesses agree to take them. If made law, it would take effect in 2014.

Democrats oppose the measure. They say it would be a bureaucratic nightmare because businesses don't have the equipment to determine the value of gold and silver.

Bureaucratic Nightmare?

Nonsense.

The bill is well written and extremely well thought out. It does not force companies to accept gold or silver (nor should it), it merely allows businesses to do so if they want. Any company that does not want to deal with gold or silver will not have to. So where's the nightmare?

States will not be minting their own money under such a proposal (nor should they) so there is no conflict on that part of Federal law.

I commend this bill, expect Arizona lawmakers to pass it, and urge the Governor to sign it. When that happens, gold will once again be legal money.

I support gold as money and believe gold is money whether or not the bill passes.

There is significant reason for people to distrust government-sponsored fiat currencies backed by nothing. I made the case recently in Fraudulent Guarantees; Fictional Reserve Lending; Comparison of US to Cyprus; What About New Zealand?

Here is a brief synopsis, but I encourage you to read the full article.

Monetary Recap

  • Base Money Supply: $2.9 Trillion
  • M1: 2.4 Trillion
  • M2: 10.4 Trillion
  • Total Credit Market Debt Owed: $56.3 trillion
One Giant Ponzi Scheme

Clearly far more money has been lent than exists. How can it possibly be paid back? If it can't be paid back, how good is a government guarantee on deposits?

In 2010 Bernanke proposed ending reserve requirements completely, but long-time Mish readers understand what Bernanke proposed is the de facto state of affairs already. (see the above link for an explanation).

Five Key Points
  1. In a Fractional Reserve Lending scheme, the notion there are meaningful reserves is ridiculous.
  2. Far more money has been lent out than really exists (the rest is a fictional accounting entry).
  3. Fractional reserve lending constitutes fraud (just as lending something you do not own is fraud).
  4. There is no way for all this money to be paid back (so it won't be).
  5. The Reserve Bank of New Zealand has the most sensible policy on deposit insurance of all the world's central banks. (NZ offers no deposit insurance). See my article for a full explanation.

In the sake of full disclosure, I own gold, silver, platinum, as well as shares in various mining corporations.

3 of 4 reptiles stolen from Calif. museum found

FRESNO, Calif. (AP) ? Three of four reptiles that were swiped from a science museum and thrown together into a garbage bag during a heist were recovered Friday, though a 3-foot-long ball python remained at large, officials said.

The 3?-foot savannah monitor lizard, a 7-foot-long red-tailed boa constrictor and another 3-foot-long ball python were in good condition, said Mary Ellen Wright, the director of the Fresno Discovery Center. Authorities were searching for the missing python in an area frequented by a suspect, who has been arrested.

Wright had been worried about the reptiles' confinement together in the bag "because they are mortal enemies," she said.

"It would be like throwing two pit bulls in a locked room," she added, noting that the monitor lizard has sharp, 2-inch claws.

The reptiles were taken in a robbery that has perplexed museum officials and authorities, who have not released a motive.

Fresno Police Lt. Donald Gross said Friday Devin Michael Madej, 20, was arrested on suspicion of burglary and possession of stolen property.

The museum's education coordinator, Ian Goudelock, said the burglar didn't appear to be out to intentionally hurt the animals though the museum planned to have the reptiles checked by a veterinarian.

"It's just a strange theft. We're still trying to figure out why," he said. "It does kind of more or less have a happy ending."

Surveillance video showed the burglar broke into the Central California museum sometime late Wednesday or early Thursday, smashed the tanks that held the four reptiles ? worth hundreds of dollars ? and popped them into a garbage bag.

He also went into the center's gift shop and stole children's toys, the phone system and the security monitor, the Fresno Bee reported.

Researchers show stem cell fate depends on 'grip'

Friday, March 29, 2013

The field of regenerative medicine holds great promise, propelled by greater understanding of how stem cells differentiate themselves into many of the body's different cell types. But clinical applications in the field have been slow to materialize, partially owing to difficulties in replicating the conditions these cells naturally experience.

A team of researchers from the University of Pennsylvania has generated new insight on how a stem cell's environment influences what type of cell a stem cell will become. They have shown that whether human mesenchymal stem cells turn into fat or bone cells depends partially on how well they can "grip" the material they are growing in.

The research was conducted by graduate student Sudhir Khetan and associate professor Jason Burdick, along with professor Christopher Chen, all of the School of Engineering and Applied Science's Department of Bioengineering. Others involved in the study include Murat Guvendiren, Wesley Legant and Daniel Cohen.

Their study was published in the journal Nature Materials.

Much research has been done on how stem cells grow on two-dimensional substrates, but comparatively little work has been done in three dimensions. Three-dimensional environments, or matrices, for stems cells have mostly been treated as simple scaffolding, rather than as a signal that influences the cells' development.

Burdick and his colleagues were interested in how these three-dimensional matrices impact mechanotransduction, which is how the cell takes information about its physical environment and translates that to chemical signaling.

"We're trying to understand how material signals can dictate stem cell response," Burdick said. "Rather than considering the material as an inert structure, it's really guiding stem cell fate and differentiation ? what kind of cells they will turn into."

The mesenchymal stem cells the researchers studied are found in bone marrow and can develop into several cell types: osteoblasts, which are found in bone; chondrocytes, which are found in cartilage; and adipocytes, which are found in fat.

The researchers cultured them in water-swollen polymer networks known as hydrogels, which share some similarities with the environments stem cells naturally grow in. These materials are generally soft and flexible ? contact lenses, for example, are a type of hydrogel ? but can vary in density and stiffness depending on the type and quantity of the bonds between the polymers. In this case, the researchers used covalently cross-linked gels, which contain irreversible chemical bonds.

When seeded on top of two-dimensional covalently cross-linked gels, mesenchymal stem cells spread and pulled on the material differently depending on how stiff it was. Critically, the mechanics guide cell fate, or the type of cells they differentiate it into. A softer environment would produce more fat-like cells and a stiffer environment, where the cells can pull on the gel harder, would produce more bone-like cells.

However, when the researchers put mesenchymal stem cells inside three-dimensional hydrogels of varying stiffness, they didn't see these kinds of changes.

"In most covalently cross-linked gels, the cells can't spread into the matrix because they can't degrade the bonds ? they all become fat cells," Burdick said. "That tells us that in 3D covalent gels the cells don't translate the mechanical information the same way they do in a 2D system."

To test this, the researchers changed the chemistry of their hydrogels so that the polymer chains were connected by a peptide that the cells could naturally degrade. They hypothesized that, as the cells spread, they would be able to get a better grip on their surrounding environment and thus be more likely to turn into bone-like cells.

In order to determine how well the cells were pulling on their environment, the researchers used a technique developed by Chen's lab called 3D traction force microscopy. This technique involves seeding the gel with microscopic beads, then tracking their location before and after a cell is removed.

"Because the gel is elastic and will relax back into its original position when you remove the cells," Chen said, "you can quantify how much the cells are pulling on the gel based on how much and which way it springs back after the cell is removed."

The results showed that the stem cells' differentiation into bone-like cells was aided by their ability to better anchor themselves into the growth environment.

"With our original experiment, we observed that the cells essentially didn't pull on the gel. They adhered to it and were viable, but we did not see bead displacement. They couldn't get a grip," Burdick said. "When we put the cells into a gel where they could degrade the bonds, we saw them spread into the matrix and deform it, displacing the beads."

As an additional test, the researchers synthesized another hydrogel. This one had the same covalent bonds that the stem cells could naturally degrade and spread through but also another type of bond that could form when exposed to light. They let the stem cells spread as before, but at the point the cells would begin to differentiate ? about a week after they were first encapsulated ? the researchers further "set" the gel by exposing it to light, forming new bonds the cells couldn't degrade.

"When we introduced these cross-links so they could no longer degrade the matrix, we saw an increase toward fat-like cells, even after letting them spread," Burdick said. "This further supports the idea that continuous degradation is needed for the cells to sense the material properties of their environment and transduce that into differentiation signals."

Burdick and his colleagues see these results as helping develop a better fundamental understanding of how to engineer tissues using stem cells.

"This is a model system for showing how the microenvironment can influence the fate of the cells," Burdick said.

HBT: D-backs sign 1B Goldschmidt to extension

No details are known yet, but Jim Bowden breaks the news?that the Diamondbacks and first baseman Paul Goldschmidt have agreed to a contract extension.

2012 was his first full season in the bigs, and he showed he belonged, hitting .286/.359/.490 with 20 home runs, 82 RBI, 18 stolen bases and an .850 OPS. While he isn?t arbitration-eligible until after the 2014 season and while he is under team control though 2017, the Diamondbacks obviously wanted to leave nothing to chance with him.

GOP moves to plug technology gap with Democrats

WASHINGTON (AP) ? Embarrassed by how the last presidential election exposed their yesteryear technology, Republicans are turning to a younger generation of tech-savvy social media experts and software designers to improve communications with voters, predict their behavior and track opponents more vigorously.

After watching President Barack Obama win re-election with help from a technology operation unprecedented in its sophistication, GOP officials concede an urgent need for catch up.

"I think everybody realized that the party is really far behind at the moment and they're doing everything within their realistic sphere of influence to catch up," said Bret Jacobson, a partner with Red Edge, a Virginia-based digital advocacy firm that represents the Republican Governors Association, the U.S. Chamber of Commerce and the Heritage Foundation.

Democrats began using related technology years ago, giving Obama a significant advantage last fall in customizing personalized fundraising and get-out-the-vote appeals to prospective supporters. With the blessing of party leaders, a new crop of Republican-backed outside groups is developing tools to do the same in 2014 and 2016.

Alex Skatell, former digital director for the GOP's gubernatorial and Senate campaign operations, leads one new group that has been quietly testing a system that would allow Republicans to share details about millions of voters ? their personal interests, group affiliations and even where they went to school.

With no primary opponent last year, Obama's re-election team used the extra time to build a large campaign operation melding a grass-roots army of 2.2 million volunteers with groundbreaking technology to target voters. They tapped about 17 million email subscribers to raise nearly $700 million online.

Data-driven analytics enabled the campaign to run daily simulations to handicap battleground states, analyze demographic trends and test alternatives for reaching voters online.

Republican presidential nominee Mitt Romney, in contrast, had only a few months after a lengthy primary fight to try to match Obama's tech advantage. He couldn't make up the difference. Romney's technology operation was overwhelmed by the intense flow of data and temporarily crashed on Election Day.

A 100-page report on how to rebound from the 2012 election, released last week by Republican Party Chairman Reince Priebus, includes several technology recommendations.

"The president's campaign significantly changed the makeup of the national electorate and identified, persuaded and turned out low-propensity voters by unleashing a barrage of human and technological resources previously unseen in a presidential contest," the report said. "Marrying grass-roots politics with technology and analytics, they successfully contacted, persuaded and turned out their margin of victory. There are many lessons to be learned from their efforts, particularly with respect to voter contact."

Skatell, 26, is leading one new effort by Republican allies to fill the void. His team of designers, software developers and veteran Republican strategists is now testing what he calls an "almost an eHarmony for matching volunteers with persuadable voters" that would let campaigns across the country share details in real time on voter preferences, harnessing social media like Facebook and Twitter.

Other groups are working to improve the GOP's data and digital performance.

The major Republican ally, American Crossroads, which spent a combined $175 million on the last election with its sister organization, hosted private meetings last month focused on data and technology. Drawing from technology experts in Silicon Valley, the organization helped craft a series of recommendations expected to be rolled out later this year.

"A good action plan that fixes our deficiencies and identifies new opportunities can help us regain our advantage within a cycle or two," said Crossroads spokesman Jonathan Collegio.

A prominent group of Republican aides has also formed America Rising, a company that will have a companion "super" political action committee that can raise unlimited contributions without having to disclose its donors. Its purpose is to counter Democratic opposition research groups, which generated negative coverage of Romney and GOP candidates last year.

America Rising will provide video tracking, opposition research and rapid response for campaign committees, super PACs and individual candidates' campaigns but does not plan to get involved in GOP primaries. It will be led by Matt Rhoades, who served as Romney's campaign manager, and Joe Pounder, the research director for the Republican National Committee. Running its super PAC will be Tim Miller, a former RNC aide and spokesman for former GOP presidential candidate Jon Huntsman.

Romney and several Republican candidates were monitored closely by camera-toting Democratic aides during the campaign, a gap that Miller said American Rising hopes to fill on behalf of Republicans.

Brad Woodhouse, a spokesman for the Democratic National Committee, said his party has "a several years' lead on data and analytics infrastructure and we're not standing still."

Of the GOP effort, Woodhouse said, "We don't see them closing the gap anytime soon."

Canadian Pacific Railway says line reopened after oil spill

(Reuters) - Canadian Pacific Railway Ltd reopened a rail line in western Minnesota on Thursday, a day after a mile-long train hauling crude oil from Alberta, Canada, derailed in the area, spilling up to 30,000 gallons of oil.

"The rail line was formally re-opened early Thursday morning, following full track repairs and mandatory inspections," said Canadian Pacific spokesman Ed Greenberg in an email.

Greenberg said the clean-up near the town of Parkers Prairie, 150 miles northwest of Minneapolis, is progressing. He added the investigation into the cause of the derailment is ongoing.

Streaking Penguins winning with defense

PITTSBURGH (AP) ? Look out, the NHL's highest scoring team is learning to play defense too.

The Pittsburgh Penguins have ripped off 13 straight wins, the second longest streak in franchise history. While they're getting the typical production from star center Sidney Crosby, the Penguins are developing into one of the league's top defensive teams.

Pittsburgh has allowed just nine goals in its last nine games, the lowest total in the NHL over that stretch. Defensesman Brooks Orpik says the team just got tired of trying to win every game in a shootout and knew it needed to tighten things up if it wanted to make a serious run at the Stanley Cup.

The Penguins will try to make it 14 in a row Thursday when they host Winnipeg.

Nintendo video shows off Wii U speed improvement coming in April update

Nintendo's Wii U has faced complaints over slow loading and switching between menus since launch, but the company has promised a pair of updates will help the situation. Tonight it posted a video on YouTube (embedded after the break) that shows off the difference before and after the April update side by side. Showing off how quickly it can return to the home menu from a game of New Super Mario Bros. U, the updated console is ready to go in eight seconds, compared to the current software's 20-second delay. There's no mention of the other update to improve the speed of launching software, but hopefully that will be shown off soon as well. More than halving the main menu's load time is nothing to sneeze at, although it's still not exactly a snappy experience. We'll see if these tweaks -- once they arrive -- do anything to improve the console's position while it waits for the improved software lineup President Satoru Iwata is expecting.

Arizona Legislature overhauls unemployment claims

PHOENIX (AP) -- A bill that would make it more difficult for recently unemployed people in Arizona to collect unemployment benefits is likely to land on Republican Gov. Jan Brewer's desk.

The state House approved legislation Tuesday that would require unemployed workers to present documents showing they were fired before they can receive benefits. Under current law, the burden is on employers to fight fraudulent claims.

The 34-24 vote came a week after the Senate passed the proposal along party lines. Republicans control the Senate and House.

The proposed change follows complaints from business leaders about workers who walk off jobs and then file for benefits.

Democrats slammed Republicans for making it harder for struggling families to seek assistance while also doing little to promote job creation. They said it would be nearly impossible for workers to prove they were let go because employers often don't provide documentation informing workers of their dismissal.

"They are already going through a hard enough time as it is," said Democratic Rep. Jonathan Larkin.

Minority Leader Chad Campbell said workers might not be able to prove they were fired if the company they worked for shut its doors. He blasted Republicans for "creating more problems for the people who don't need more problems in their lives"

"Those are the people we should be helping," he said. "We should be down here trying to create jobs."

Republicans countered that the bill would create jobs because it would protect businesses from devoting hours to fighting fraudulent claims instead of filling open positions.

"They are begging for workers," said Republican Rep. Steve Montenegro. "They need people employed, but sometimes, unfortunately, not everybody, but there are people who chose to be on unemployment rather than take some of those jobs."

Republican Rep. Eddie Farnsworth said too often state unemployment assistance workers err on the side of jobless workers over businesses "because they have deep pockets and the person who quit has no responsibility."

"You have the responsibility to show that and not the business," Farnsworth said.

The U.S. Department of Labor initially raised concerns about the legality of the bill. Under federal law, applicants who have lost a job through no fault of their own and are able to work are eligible to receive benefits. The burden of proof is not on the employee.

"It is the state's responsibility to make reasonable efforts to obtain the information necessary to determine whether an individual is eligible," federal officials wrote to lawmakers in early March.

In the Senate, lawmakers amended the bill to address those federal concerns. The legislation now requires the state to make a final determination based on all submitted evidence.

Under the measure, employers would be allowed to claim that the employee submitted a resignation orally to prove the worker voluntarily left the job. Employers also could challenge claims if they say the worker skipped work.

Thousands of people could be impacted by the proposed change. The number of people claiming benefits in Arizona was about 75,000 in January, according to the state Department of Economic Security.

Arizona is already tougher than many other states when it comes to unemployment claims. The maximum weekly unemployment benefit in Arizona is $240. The national average has been $300 since 2010, according to the Center on Budget and Policy Priorities in Washington, D.C.

Gunman fires on homes in Tacoma, Wash.; police surround residence

By M. Alex Johnson, staff writer, NBC News

A man who reportedly walked through a Tacoma, Wash. neighborhood spraying gunfire was taken into police custody after a standoff at his residence, Seattle TV station KING 5 reported.

Police had surrounded the home Tuesday afternoon, where the man with at least four high-powered weapons was holed up, a sheriff's official told NBC News.

No injuries were immediately reported, but Pierce County sheriff's Detective Ed Troyer said several homes were hit by bullets in the neighborhood in Tacoma, about 30 miles of Seattle.

It wasn't known whether anybody else was in the home with the gunman, who Troyer said was known to have "mental health issues" and was believed to have been drinking.

Police began firing tear gas into the house early Tuesday evening, KING reported.

KING reported that numerous residents of the neighborhood had been evacuated and that those who remained were asked to stay inside.

Troyer told KING that the man, who he said was 67 years old and also had substance abuse issues, crashed his car last week in a DUI accident.

New tiger reserve is established in India

A tigress and her cub rest in India's Sathyamangalam Wildlife Sanctuary.

By Douglas Main
LiveScience

India has created a new protected area for tigers within a wildlife reserve in the south of the country.

The area, part of the Sathyamangalam Wildlife Sanctuary, is home to about 25 tigers, according to a release from the World Wildlife Fund (WWF), a conservation group. This population of tigers rivals the size of some of India's better-known reserves, the statement said.

This will be the 42nd tiger reserve in the country, which is home to the largest population of tigers?in the world. The 272-square-mile (705 square kilometers) protected area will help connect several adjacent parks, making it one of the largest continuous tiger habitats in the world, according to the WWF. The area is also home to elephants, leopards, hyenas and vultures.

For more than a decade, WWF-India has worked with local authorities in the state of Tamil Nadu (where the reserve is found) to support projects to counter poaching, improve communications via cellular phones and wireless networks, train forest rangers and monitor tigers, the statement said.

"The tiger is the national animal of India, and WWF congratulates the government for yet another important milestone in its conservation efforts that will make a tremendous contribution to the goal of conserving wild tigers and their natural habitats in the country," said Dipankar Ghose, of WWF-India, in the statement.

Tiger numbers have declined by about 95 percent in the last century across their entire historic range, and experts think there are only about 3,000 left in the wild, according to the International Union for Conservation of Nature.

The 2011 labor agreement included an important provision:? HGH testing is coming to the NFL.? Nearly 20 months later, HGH testing is no closer than it was before the agreement was signed.

The latest evidence comes from the case of Andrus Veerpalu, an Estonian skier whose three-year suspension was overturned by the Court of Arbitration for Sport.? Naturally, the NFL and the NFLPA disagree as to the meaning of the ruling, and the incident has caused Albert Breer of NFL Network to learn that the two sides have scrapped an agreement to conduct a so-called ?population study? aimed at gauging the permissible natural levels of HGH in football players.

The details don?t matter, because neither the NFL nor Congress is willing to do anything more than huff and puff about the union?s refusal to honor the agreement to submit to HGH testing.? As a result, the perception is that neither the NFLPA nor the NFL truly want HGH testing.

Since the day the NFL banned the use of HGH, the prohibition has been enforced via the honor system.? The problem with the honor system? ? It works roughly as well as the rhythm method.? So with no way to test for HGH, players will get caught only if a vial of HGH falls out of their letterman jackets, or if the player?s name pops up in the records of an HGH supplier the government is prosecuting.

Surely, the NFL and the NFLPA realize that, if/when HGH testing begins, plenty of players will be caught.? Which will reduce the supply of healthy players.? In turn, players who quit using HGH will not recover as quickly from injuries, likewise reducing the supply of healthy players.

And it won?t be good for the game if players are busted for using HGH, even though most fans presume that they?re using something to get big, to stay big, and/or to rebound from big hits applied by other big men.

If the NFL truly wanted to force the issue on HGH testing, wouldn?t the league unleash the legal hounds and push the issue in court or via an arbitration?? The players already have agreed to submit to testing, and the NFL has more than enough ammunition to argue that the NFLPA deliberately is dragging its feet.? The idea that the NFL doesn?t want to force players to the needle by court order only goes so far.? At some point, the NFL needs to do more than complain about the NFLPA?s refusal to proceed, or the NFLPA will continue to refuse to proceed.

Likewise, Congress has proven to be impotent on the topic, periodically issuing hollow threats but never taking action.Through it all, the delay has given those who use HGH an opportunity to find better masking agents ? or to develop the next wave of substances that work like HGH but for which testing doesn?t yet exist.

The best news for the NFL, the NFLPA, and Congress is that neither the media nor the fans seem to care that the NFL and the NFLPA have struck a deal to abandon the honor system, but that the honor system has continued to be used for two seasons, and counting.

Gene therapy may aid failing hearts

Mar. 25, 2013 ? In an animal study, researchers at the University of Washington show that it was possible to use gene therapy to boost heart muscle function. The finding suggests that it might be possible to use this approach to treat patients whose hearts have been weakened by heart attacks and other heart conditions.

Led by University of Washington (UW) Professor and Vice Chair of Bioengineering Michael Regnier and Dr. Chuck Murry, director of the Center for Cardiovascular Biology and co-director of the Institute for Stem Cell and Regenerative Medicine at UW, the study appears online today in the journal Proceedings of the National Academy of Sciences (PNAS).

Normally, muscle contraction is powered by a molecule, the nucleotide called Adenosine-5'-triphosphate (ATP). Other naturally occurring nucleotides can also power muscle contraction, but, in most cases, they have proven to be less effective than ATP.

In an earlier study of isolated muscle, however, Regnier, Murry and colleagues had found that one naturally occurring molecule, called 2 deoxy-ATP (dATP), was actually more effective than ATP in powering muscle contraction, increasing both the speed and force of the contraction, at least over the short-term.

In the new PNAS study, the researchers wanted to see whether this effect could be sustained. To do this, they used genetic engineering to create a strain of mice whose cells produced higher-than-normal levels of an enzyme called Ribonucleotide Reductase, which converts the precursor of ATP, adenosine-5'-diphosphate or ADP, to dADP, which, in turn, is rapidly converted to dATP.

"This fundamental discovery, that dATP can act as a 'super-fuel' for the contractile machinery of the heart, or myofilaments, opens up the possibility to treat a variety of heart failure conditions," Regnier said. "An exciting aspect of this study and our ongoing work is that a relatively small increase in dATP in the heart cells has a big effect on heart performance."

The researchers found that increased production of the enzyme Ribonucleotide Reductase increased the concentration of dATP within heart cells approximately tenfold, and even though this level was still less than one to two percent of the cell's total pool of ATP, the increase led to a sustained improvement in heart muscle function, with the genetically engineered hearts contracting more quickly and with greater force.

"It looks as though we may have stumbled on an important pathway that nature uses to regulate heart contractility," Murry added. "The same pathway that heart cells use to make the building blocks for DNA during embryonic growth makes dATP to supercharge contraction when the adult heart is mechanically stressed."

Importantly, the elevated dATP effect was achieved without imposing additional metabolic demands on the cells, suggesting the modification would not harm the cell's functioning over the long-term.

The finding, the authors write, suggest that treatments that elevate dATP levels in heart cells may prove to be an effective treatment for heart failure.

Why sticking around is sometimes the better choice for males

Mar. 26, 2013 ? Researchers from Lund University and the University of Oxford have been able to provide one answer as to why males in many species still provide paternal care, even when their offspring may not belong to them. The study finds that, when the conditions are right, sticking around despite being 'cuckolded' actually turns out to be the most successful evolutionary strategy.

The study, by Charlie Cornwallis and colleagues, is published 26 March in the open access journal PLOS Biology.

In many species, males put a lot of effort into caring for offspring that are not their own. At first glance this makes little sense, because natural selection should dictate that males only care for the offspring that carry their genes. However, this study suggests that the males are both more tolerant and more astute than previously assumed, and in fact adjust their care according to how likely it is that females are unfaithful, whilst also judging whether caring will potentially reduce the number of offspring they can have in the future.

The researchers conducted a meta-analysis of 62 studies across 48 different species including insects, fish, birds and mammals. Overall, the researchers found that promiscuous copulations by females reduced the investment of males by 12%. Although parental care is highly variable across these species, the researchers were able to find a general explanation for why sticking around to care for the offspring is the better choice for some males that have been usurped. The reason is that males tend to be more accepting of offspring fathered by other males in species where the risk of cuckoldry is generally low, or when caring does not harm their future reproductive success.

"This, to me, shows the strength of natural selection, with its footprints clear in species from burying beetles -- which care for young over a few weeks by regurgitating dead mice -- to humans, who spend years providing for their children," says Charlie Cornwallis, researcher at the Department of Biology, Lund University. "These are complex calculations that males are making," he adds, "and it has been difficult to measure the relevant factors correctly, but looking across species has helped us work out what is going on. Moreover, a comparative study like this can guide researchers to the types of species and experimental cues that are likely to provide the most insight into paternal care in the future."

The study therefore opens up the possibility of more targeted research in the area. Now that the researchers know what factors are important, they can design studies to further test their findings and predict what males will do in species that have not yet been studied. For example, in species where the cost of caring is very low, males would not be expected to adjust their level of parental care even if the females are promiscuous. Rather than these males being 'duped', such tolerance has actually been favoured by natural selection.

Jordan closes main border crossing with Syria

AMMAN (Reuters) - Jordan closed its main border crossing with Syria on Monday after two days of fighting there between Syrian troops and rebel fighters, Jordan's information minister said.

Border traders said passengers were turned back at the Jordanian border crossing of Jaber and prevented from entering in the first such closure of the crossing since the revolt against President Bashar al-Assad's rule started two years ago.

"The border post is effectively closed because there have been clashes since yesterday and they are continuing," minister Samih Maaytah told Reuters.

The crossing is a thoroughfare for passengers and was transit route before the Syria conflict for hundreds of trucks daily carrying goods to Turkey from the Gulf and vice versa.

Situated near the Jordanian border, Syria's southern Deraa province is a vital battleground. Rebels in the last month have stepped up fighting for control of the border area and Deraa town after gaining territory in the countryside and capturing several army bases.

They have also overrun several towns near the Israeli-occupied Golan Heights, fuelling tensions in the sensitive military zone.

Major advance in understanding risky but effective multiple sclerosis treatment

Mar. 26, 2013 ? A new study by multiple sclerosis researchers at three? Canadian centres addresses why bone marrow transplantation (BMT) has positive results in patients with particularly aggressive forms of MS. The transplantation treatment, which is performed as part of a clinical trial and carries potentially serious risks, virtually stops all new relapsing activity as observed upon clinical examination and brain MRI scans. The study reveals how the immune system changes as a result of the transplantation. Specifically, a sub-set of T cells in the immune system known as Th17 cells, have a substantially diminished function following the treatment.

The finding to be published in the upcoming issue of Annals of Neurology and currently in the early online version, provides important insight into how and why BMT treatment works as well as how relapses may develop in MS.

"Our study examined why patients essentially stop having relapses and new brain lesions after the bone marrow transplant treatment, which involves ablative chemotherapy followed by stem cell transplantation using the patient's own cells," said Prof. Amit Bar-Or, the principle investigator of the study, who is a neurologist and MS researcher at The Montreal Neurological Institute and Hospital -The Neuro, McGill University, and Director of The Neuro's Experimental Therapeutics Program. "We discovered differences between the immune responses of these patients before and after treatment, which point to a particular type of immune response as the potential perpetrator of relapses in MS."

"Although the immune system that re-emerges in these patients from their stem cells is generally intact, we identified a selectively diminished capacity of their Th17 immune responses following therapy -- which could explain the lack of new MS disease activity. In untreated patients, these Th17 cells may be particularly important in breaching the blood-brain-barrier, which normally protects the central nervous system. This interaction of Th17 cells with the blood-brain barrier can facilitate subsequent invasion of other immune cells such as Th1 cells, which are thought to also contribute to brain cell injury.

Twenty-four patients participated in the overall clinical trial as part of the 'Canadian MS BMT' clinical trial, coordinated by Drs. Mark Freedman and Harry Atkins at the Ottawa General Hospital. The new discovery, made in a subset of patients participating in the clinical trial, was based on immunological studies carried out jointly in laboratories at The Neuro and the Universit? de Montr?al. Results of this study not only show the clinical benefits of BMT treatment, but also open a unique window into the immunological mechanisms underlying relapses in MS. Th17 cells could be the immune cells associated with the initiation of new relapsing disease activity in this group of patients with aggressive MS. This finding deepens our understanding of MS and could guide the development of personalized medicine with a more favourable risk/benefit profile.

Among the patients treated in the Canadian MS BMT clinical trial, was Dr. Alexander Normandin, a family doctor, who was a third- year McGill medical student getting ready for his surgery exams when he first learned he had MS, "I was so engrossed in my studies that I didn't pay attention to the first sign but within a few days of waking up with a numb temple, my face felt frozen. I learned that I had a very aggressive form of MS and would probably be in a wheelchair within a year. It was a brutal blow. I became patient #19 -- of only 24 for this experimental treatment. My immune system was knocked out and then rebooted with my stem cells. Today, my MS has stabilized. I now have this disease under control and I take it one day at a time."

Both the clinical and biological studies were supported by the Research Foundation of the Multiple Sclerosis Society of Canada.

Multiple Sclerosis

MS is a disorder of the brain and spinal cord that causes fatigue, disequilibrium, sensory problems and muscle paralysis. The cause of MS is unknown, but evidence suggests that it is an auto-immune disease that destroys myelin, a substance coating axons, the thin strands that carry signals between brain cells. It usually strikes between the ages of 15 and 40 but can begin as early as age two. Women have twice the probability of developing MS than men. Canada has one of the world's highest national rates -- about 1,100 new cases each year. Some 50,000 Canadians have MS. More than 1 in 5 lives in Quebec. The most common form of MS is relapsing-remitting, in which acute symptoms alternate with periods of remission. Primary progressive MS, the least common form, develops continually without remission. Secondary progressive MS begins as relapsing-remitting, then becomes steadily progressive.

Google selects Glass Explorers, will send invites to winners over the next few days

What would you do if you had Glass? Google asked you to share your plans last month, offering up 8,000 coveted reservations in the search giant's "Explorers" program. Now, the company has finally selected the winners, and plans to send out order invitations "over the next few days." Sadly, invite recipients will still need to hand over $1,500 (plus tax) for their very own set, and the program is currently limited to folks in the US, but this was still the best opportunity to get an early look at Glass, making it a very appealing program, despite the cost. Google received many creative applications, including a proposal from JetBlue, but only individual users will have an opportunity to make a purchase this time around -- the company says that it's "working on connecting with businesses in other ways." Congratulations to the lucky winners -- feel free to send us a tip when word finally hits your Google+ or Twitter inbox.

Prince Harry to tour Hurricane Sandy damage in US visit

Chris Jackson / Getty Images

By Eun Kyung Kim, TODAY contributor

Prince Harry plans to visit the United States on an official multi-stop journey that will include a visit to wounded veterans.

The prince will be in America from May 9-15, the palace said in a briefing Monday. His trip will include a stop in Washington, D.C., where he will visit the Walter Reed National Military Medical Center, a facility for wounded service members.

Harry, an officer in the British Army, recently returned from a second tour of duty in Afghanistan.

The prince also plans to visit a Capitol Hill exhibit on landmine clearance, a campaign passionately taken up by his mother, Princess Diana.

Harry also will tour areas hit by Hurricane Sandy in New York and New Jersey, along with a visit to Greenwich, Conn., to attend a polo match. Denver also will be on his tour stop.

The May trip will be an official visit on behalf of England, unlike the last time the prince visited the United States in August 2012 following the Summer Olympics. He spent several days in Las Vegas, where he was famously photographed nude by friends he met at the time. Harry later said he showed poor judgment, and that ?I let my family down; I let other people down? with the incident.

LinkedIn gets new Facebook-like search for jobs, companies, people

LinkedIn?s new Unified Search is interesting, convenient, and a pleasure to use. It?s also set up a lot like Facebook?s recently released Graph Search, with lots of autocomplete functionality and results that combine many kinds of information into a single personalized list of results.

?The results are always influenced by the composition of my network, who I?m connected to as well as who I am and what?s in my profile,? said Brad Mauney, LinkedIn product lead for identity products, in a recent interview with VentureBeat.

In order to make the new Unified Search, as it?s called, as smart as possible, the team behind it considered content (like posts and groups) as well as context (who you?re connected to and what tasks you?re trying to accomplish with each search.

The result is an all-new search feature that shows all results grouped by companies, people, groups, posted jobs, updates, etc. It even allows you to search your LinkedIn inbox as part of the new, personalized search experience. All these different types of results are returned any time you start a new search; you can choose how to narrow it down.

You can also do a highly specific people search ? perfect for finding those two or three or five leads who can get you exactly what you want, whether it?s a big sale or a new job ? and you can save searches and get alerts when new results come up on LinkedIn?s radar.

Here?s a sneak peek:

?We think of identity as finding, connecting, and being found, and search is obviously part of that,? said Mauney. ?Search has always been a core part of LinkedIn?s DNA ? and it?s always been a social experience.?

As far as the product?s resemblance to Graph Search goes, it?s obvious the two teams (at Facebook and LinkedIn) were working separately but in parallel and that a more object-oriented approach to search (as opposed to a keyword/string-based search) is going to become the new normal.

?The way we?re going with search is very much intended to let our members navigate through a very complex system of things and relationships and be effective as professionals in terms of finding new opportunities, new candidates, new jobs, advice,? said Mauney.

?No two professionals are alike, and no two users on LinkedIn are alike. The graph aspect here and that we?re seeing evolving is intended to capitalize on that degree of personalization.?

LinkedIn also hopes to highlight some of the more active and exciting parts of its site and service, like its massive database of groups posts and its daily news sections.

?If you look at the direction we?ve been going recently, the products we?ve invested in like LinkedIn Today, LinkedIn News, and the influencer program, the quality continues to amaze me,? Mauney said.

?Imagine a LinkedIn where you go to the search box and search for entrepreneurship, and we know you?re looking for content and groups, and we want to drive you to the place where you?ll be most engaged.?

Wouldn?t you typically look to Google for a search on a wide-open topic like that? I ask.

?If you?re looking for the page-ranked version of ?best,? you go to Google,? Mauney explained.

?If you want to see what?s resonating with your network, if your LinkedIn network is good, LinkedIn is going to surface the best content. ? Using your professional graph as a primary filter against the firehose of news and content that we?re being blasted with every day ? it?s a very powerful thing.?
